Study on nanodelivery system based on active and precise regulation of protein crowns

The nanodelivery system based on the active and precise regulation of protein crown is a new strategy that uses the protein crown (protein corona) on the surface of nanomaterials as a regulatory factor to realize the payload, protection and release of drugs by changing the characteristics, structure and function of nanomaterials. Protein crown refers to a layer of covering formed by non-specific interaction with biomolecules (mainly proteins) after entering a biological organism, which can affect the biological behavior and effects of nanomaterials. The nanodelivery system based on the active and precise regulation of protein crowns can make use of the diversity and controllability of protein crowns to realize the adaptive regulation of the tumor microenvironment, so as to improve the targeting and bioavailability of drugs, reduce toxic and side effects, and enhance the therapeutic effect.

A nanodelivery system based on GO nanosheet (GO) and polyethylene glycol (PEG), which can responsive change the PEG density of GO surface through pH, so as to realize the dynamic regulation of protein crown, and then affect the GO transport and drug release in tumor cells. The authors demonstrated the good biosafety, stability, and targeting of this nanodelivery system and demonstrated its excellent antitumor effect in vitro and in vivo.
