Protein L is derived from Peptostreptococcus magnus. It is a protein that can bind to a wide range of immunoglobulins. The protein only binds to the κ light chain of the antibody without affecting the antigen binding site of the antibody. One feature makes it more widely combined with antibodies from various sources and subclasses compared with protein A or protein G. It does not bind to immunoglobulins derived from cattle, goats or sheep, and can be used to purify monoclonal antibodies from culture broth supplemented with BSA or FCS. The natural protein L is composed of 719 amino acids, does not contain disulfide bonds, and is an acidic molecule PI 4. It binds to the light chain (L chain) of IgG. Protein L mainly has affinity with Kappa light chain. Protein L has affinity for different IgG, IgM, IgA, IgE and IgD. Protein L binds to the Kappa region of the light chain and has no affinity with lambda. The affinity sites are in the I, III, and IV regions of the light chain.






Human immunoglobulin light chain contains two subtypes, Lambda and Kappa. About 60% of IgG is kappa and 40% of IgG contains Lambda. Before establishing a purification method, it is best to identify the type of light chain and select the correct chromatographic medium for separation and purification.
